Minocycline hydrochloride is a semisynthetic derivative of tetracycline. It is primarily bacteriostatic and acts by inhibiting protein synthesis. It has a similar antimicrobial spectrum of activity against a wide range of gram-positive and gram-negative organisms as other tetracyclines.

Excretion:
Fecal and Renal: one-half to one-third that of other tetracycline
Dialyzable: no (hemodialysis), no (peritoneal dialysis)
Elimination Half Life:
(Oral), 15.5 h (11.1 h to 22.1 h)
Patients with hepatic dysfunction: 11 h to 16 h
Patients with renal dysfunction: 18 h to 69 h .

Hypersensitivity to any tetracycline or any component of the product.

Fetal risk has been demonstrated. (TH)

Common:
Gastrointestinal: Tooth discolored
Neurologic: Dizziness (9% ), Headache (23% )
Other: Fatigue (9% )
Serious:
Dermatologic: Erythema multiforme, Stevens-Johnson syndrome
Gastrointestinal: Clostridium difficile colitis
Hepatic: Hepatic failure, acute, Hepatitis
Immunologic: Anaphylaxis, Drug reaction with eosinophilia and systemic symptoms, Hypersensitivity reaction, Systemic lupus erythematosus
Neurologic: Bulging fontanelle, Pseudotumor cerebri
Other: Serum sickness due to drug.

Initial: 200 mg for 1 dose; Maintenance: 100 mg every 12 hours; more frequent dosing intervals may be used (100 to 200 mg initially, followed by 50 mg 4 times daily).(UTD)

Children >8 years and Adolescents: Initial: 4 mg/kg/dose for 1 dose; Maintenance: 2 mg/kg/dose every 12 hours (maximum: 400 mg daily)(UTD)

CrCl ?80 mL/minute: No dosage adjustment necessary.
CrCl <80 mL/minute: Do not exceed 200 mg daily.(UTD)

There are no dosage adjustments provided in the manufacturer's labeling; however, hepatotoxicity has been reported. Use with caution.(UTD)
