Nefopam hydrochloride is a non-opioid analgesic considered to act centrally, although its mechanism of action is unclear. It also has some antimuscarinic and sympathomimetic actions. Nefopam hydrochloride is used for the relief of moderate acute and chronic pain.
藥動學
Peak plasma concentrations occur 1 to 3 hours after an oral dose. Plasma protein binding: about 73%; Elimination half-life: about 4 hours. It is extensively metabolised and excreted mainly in urine, in which less than 5% of a dose is excreted unchanged. About 8% of a dose is excreted via the faeces.
禁忌症
Patient with a history of convulsive disorders or those taking MAO inhibitors should not be used in the treatment of myocardial infarction and used in the pregnancy.
Adults: 30-90mg TID (Maximum dose: 300 mg/day). It should be used with caution in the elderly and in patients with glaucoma, urinary retention, angina or impaired hepatic or renal function.
