Antipsychotic, mood elevating; antiemetic, anti-gastrin secretion

Sulpiride is a selective dopamine D2 antagonist with antipsychotic and antidepressant activity.

Sulpiride is absorbed slowly from the gastrointestinal tract. Its oral bioavailability is only 25 to 35% with marked interindividual differences. Half-life: 6-8hr.

1. Hypersensitivity to Sulpiride
2. Pheochromocytoma
3. Parkinson's disease

Sulpiride should not be used during pregnancy, especially during the last trimester. If used in 3rd trimester, extrapyramidal effects in neonates reported.

Excreted in milk in significant amounts. Possible adverse effects on developing nervous systems of infants exposed. Should be avoided unless absolutely necessary.

Anti-gastrin secretion:
1. Endocrine system: Gynecomastia, hyperprolactinemia
2. Extra-pyramidal reactions
3. Gastrointestinal: nausea, vomit, constipation
4. Others: insomnia, dizziness, drowsiness
Antipsychotic:
1. Cardionascular: abnormal blood pressure, chest tightness, tachycardia
2. Neuroleptic malignant syndrome
3. Extra-pyramidal reactions: Parkinsonism, dyskinesia, akathisia
4. Endocrine system: Gynecomastia, hyperprolactinemia
5. Others: cholestatic jaundice, sleep disturbances, over-stimulation, agitation.

Schizophrenia:
300-600 mg daily, Maximum daily dose 1200mg/day
Depression:
150-300 mg daily, Maximum daily dose 600mg/day
Anti-gastrin secretion:
150 mg daily, divided into three times
