胃癌、胰臟癌
Gastric cancer:
Treatment of disseminated adenocarcinoma of the stomach (in combination with other chemotherapy agents) and as palliative treatment when other modalities have failed.
Pancreatic cancer:
Treatment of disseminated adenocarcinoma of the pancreas (in combination with other chemotherapy agents) and as palliative treatment when other modalities have failed.
* Limitations of use: Not recommended for single-agent primary therapy or to replace appropriate surgery and/or radiotherapy in the treatment of these conditions.

Antineoplastic agents, DNA alkylator
Mitomycin alkylates DNA to produce DNA cross-linking (primarily with guanine and cytosine pairs) and inhibits DNA and RNA synthesis. Mitomycin is not cell cycle specific but has its maximum effect against cells in late G and early S phases.

1. Metabolism: Primarily hepatic
2. Half-life elimination: 17 minutes (30 mg dose)
3. Excretion: Feces; Urine

1. Hypersensitivity to mitomycin or any component of the formulation
2. Thrombocytopenia; Coagulation disorders,or other increased bleeding tendency

Adverse events have been observed in animal reproduction studies.

1.It is not known if mitomycin is present in breast milk.
2.Due to the potential for serious adverse reactions in the breastfed infant, breastfeeding is not recommended by the manufacturer.

Common:
1. Gastrointestinal: Anorexia, nausea, vomiting
2. Hematologic & oncologic: Bone marrow depression, hemolytic-uremic syndrome, thrombotic thrombocytopenic purpura
3. Miscellaneous: Fever
Postmarketing:
Adult respiratory distress syndrome (ARDS), bladder spasm, cardiac failure, dyspnea, extravasation reactions, hepatic sinusoidal obstruction syndrome, interstitial fibrosis, malaise, nonproductive cough, pulmonary infiltrates, renal failure, skin rash, weakness

IV 20mg/m2 Q6~8W

Refer to adult dosing.

1. CrCl <10 mL/minute: Reduce dose to 75% of usual dose.
2. Continuous ambulatory peritoneal dialysis (CAPD): Reduce dose to 75% of usual dose.

There are no dosage adjustments provided in the manufacturer's labeling (has not been studied).
